Reconstituting Lyophilized Research Peptides

Lyophilized (freeze-dried) peptides are shipped as a dry solid for stability and must be reconstituted into solution before use in research. This reference outlines the general laboratory standards researchers follow when bringing a research-grade peptide into solution. It is methodology reference for in-vitro laboratory research only and is not guidance for human or veterinary use.

Choosing a Diluent

Bacteriostatic water (water containing 0.9% benzyl alcohol) is the most common diluent for research reconstitution because the preservative limits microbial growth across repeated withdrawals from a multi-use vial. Sterile or ultrapure water is used where a preservative-free solution is required by the protocol. The choice of diluent is a documented part of the experimental method.

Sterile Technique

Reconstitution is performed using clean, sterile technique: the diluent is added slowly down the inner wall of the vial rather than directly onto the lyophilized pellet, and the vial is gently swirled rather than shaken to avoid mechanical stress on the peptide. The solid is allowed to dissolve fully before the solution is used.

Aliquoting and Storage

For research use, reconstituted peptide is commonly divided into single-use aliquots to avoid repeated freeze-thaw cycles, which can degrade peptide integrity over time. Aliquots are typically stored frozen (commonly at -20°C or lower for longer-term storage), while a working vial may be held refrigerated for short periods. Lyophilized material that has not yet been reconstituted is the most stable form and is kept cold and dry until use.
